Prognostic significance of the assessment of the expression of immunohistochemical markers and the degree of pathomorphism before and after treatment of patients with prostate cancer at high risk of progression

Summary:

Purpose: to analyze the expression of immunohistochemical markers in patients with prostate adenocarcinoma at high risk of progression before and after combined and combined treatment of patients with prostate specific antigen (PSA) level of blood below discriminatory. Material and methods. Examined patients (n = 80) with a diagnosis of “High-risk progression prostate cancer,” with clinical stage T3a, total score for Gleason scale from 8 to 10, the level of total PSA more than 20 ng / ml. Patients of the 1st group (n = 42) only combined surgical treatment in the scope of TUR and HIFU ablation the prostate; The 2nd group of patients (n = 38) additionally applied androgen deprivation in adjuvant mode. Results. Blood PSA, determined in patients after various methods of treatment is not always an objective indicator of its effectiveness. Immunohistochemical studies of tumor tissue for proliferative activity tumor cells, the activation of apoptosis in them, as well as the assessment of specific tumor markers and tumor suppressor genes showed that a decrease in the expression of these markers in patients with PSA values ​​of blood lower than discriminatory observed in all patients independently on the type of treatment. However, after treatment in the 1st group of patients, despite the achievement PSA level of nadir, there is a decrease in the expression of an intercellular adhesion marker E-cadherin, which indicates an increased risk of tumor metastasis, as well as increased expression of markers of the apoptosis inhibitor Bcl-2 and androgens, which indicates about the possibility of progression of tumor growth. Conclusion For a more accurate assessment therapeutic pathomorphism in patients with prostate cancer at high risk of progression, which HIFU ablation monotherapy was carried out, along with the determination of blood PSA level, it is necessary during the control biopsy of the prostate immunohistochemical studies of tumor tissue to determine the prognosis and risk of relapse.

Цель: оценить воздействие однократного внутривенного введения покрытых полиэтиленгликолем золотых наностержней на молекулярные маркеры межклеточного взаимодействия и показатели перекисного окисления в сыворотке крови четырех групп крыс: интактных, с аллоксановым диабетом, с перевитым раком печени; с перевитым раком печени на фоне аллоксанового диабета. Материалы и методы. Активность перекисного окисления липидов оценивалась по показателям гидроперекисей липидов, малонового диальдегида и молекул средней массы с использованием общепринятых спектрофотометрических методов, исследования молекулярных маркеров — фактора некроза опухоли TNF-a, инсулиноподобного фактора роста IGF-1, васкулоэн-дотелиального фактора роста VEGF-C проводились в сыворотке крови методом иммуноферментного анализа. Результаты. Выявлено, что при однократном внутривенном введении золотых наностержней не происходит значительного изменения количества маркеров межклеточного взаимодействия и показателей перекисного окисления липидов. в периферической крови. Заключение. Полученные результаты позволяют рекомендовать использование покрытых полиэтиленгликолем золотых наностержней в экспериментальной онкологии.

Summary:

Aim: to evaluate the severity of morphological changes in the liver of laboratory animals with prolonged intake of pegilated gold nanoparticles of different sizes from 2 to 50 nm. Material and methods. The experiment was performed on 240 albino rats divided into 4 groups. The animals of experimental groups were orally administered the gold nanoparticles in a specific pattern. Results. Morphological changes in liver depend on the particle size and the duration of administration. Developing pathological changes in the liver are reversible, as evidenced by the gradual recovery of the liver structure 14 days after administration. Conclusion. The most significant changes in the liver were found in the experimental groups with a 30-day administration of 2 and 50 nm gold nanoparticles that show the size-dependent nature of their impact.

Summary:

Laser thermal therapy is increasingly being used in clinical practice for cancer treatment. Its major limitation is associated with low spatial selectivity. This restriction may be excluded by the use of plasmon-resonant gold nanoparticles as photothermal sensitizers. Tunable synthesis, unique optical properties and low toxicity make gold nanostructures promising as a therapeutic agent for cancer treatment. The research work has presented the experimental data that concern the use of plasmon photothermal therapy for the treatment of transplanted and induced tumors of laboratory animals. Taking into consideration the research results it is determined that the transition of this therapeutic technology into the clinical practice can be predicted.

Summary:

The purpose of the work is to study antitumor activity of flavonoid-containing extracts of anthocyan maize and gratiola in experiments in vivo and to estimate their effects on the internal organs, tumors and blood of rats with transplanted liver cancer PC-1. Materials and methods: Effect of extracts of anthocyan maize and gratiola has been studied in experiments in vivo in 30 laboratory rats with transplanted liver cancer PC-1 using morphological and biochemical methods. Results: It has been found that extract of gratiola received by a certain way has selective cytotoxic effect on tumor cells. Anthocyan maize extract reduces endogenous intoxication. Conclusion: These results open the prospect of further study of the extract of gratiola in various experimental models of cancer and anthocyan maize extract use may be developed as a means of endogenous intoxication reducing for application in the treatment of cancer. Investigation of anthocyan maize extract may assist in the development of drugs that reduce the endogenous intoxication at the treatment of cancer.

Summary:

The purpose of the article is to assess the severity of the morphological changes in the internal organs of laboratory animals with prolonged oral administration of gold nanoparticles of different sizes. Material and methods: The experiment was carried out on 40 albino rats which were divided into 4 groups. Animals of the experimental group received oral gold nanoparticles by the appropriate scheme. Results: At comparing the morphological changes in the internal organs of laboratory animals it was established that the introduction of gold nanoparticles of different sizes leads to the same type of disturbances of morphological structure, the gold nanoparticles of 1-3 and 50 nm cause the most expressive changes. Conclusion: The most significant morphological modifications in the internal organs were caused by gold nanoparticles of 1-3 and 50 nm. It points out to the size-dependence of the effect of gold nanoparticles.

Summary:

The aim of the article is to conduct analysis of accumulation and biodistribution of gold nanoparticles in the structural and functional areas of mesenteric lymph nodes of white rats. Particular attention is paid to the size and duration of oral administration. Material and Methods: Dark field microscopy with Leica DM 2500 microscope has been used to assess biodistribution of gold nanoparticle. The histochemical reaction of silver amplification with silver lactate by Danscher has been used to confirm gold nanoparticles identification. Results: It has been found that regardless of the duration of administration gold nanoparticles of 1-3 nm have not been detected by these methods in the lymph nodes. Gold nanoparticles of 15 and 50 nm have been found out in the form of clusters in the cytoplasm of macrophages and lymphocytes mainly in the mantle zone of lymphoid follicles and brain strands. Conclusion: It has been established that the gold nanoparticles of 15 and 50 nm has been detected in the form of clusters in the cytoplasm of macrophages and lymphocytes.

Summary:

The research goal is to determine clinical, endoscopic and morphological signs of chronic duodenitis in children. Materials and methods: The diagnostic value of molecular markers has been revealed by immunohistochemical research of biopsy. It has been received from endoscopic examination of 32 children aged from 3 to 17 years old with chronic duodenitis. Morphometric investigation of markers expression has been carried out by means of analyzing system of digital images of Mikrovizor medical uVizo-103. Results: The index of proliferation has been higher in cells of cover epithelium than in glands. The inductor expression of apoptosis Bax has been poorly expressed in both groups of patients. It has been proved that proliferative processes are predominant in this pathology. Conclusion: It has been found out that proliferation of significant activity in the cover epithelium accompanies the development of preatrophic processes in children with chronic duodenitis. The greater degree of regeneration has been marked in crypts.

This review is devoted to a comparative analysis of receptor status, immunity, angiogenesis, metastatic mammal glands cancer expression profiling in humans and animals. Angiogenesis has been assessed by quantitative and immunohistochemical characteristics by means of evaluation of microvascular density (MVD) using Claudin-5 (CLDN-5) as a marker of vascular endothelium in tumors of mammary glands in dogs.
