Investigation of the relationship between changes in thermographic and flowmetric parameters of skin peripheral hemodynamics in laboratory rats

Summary:

Purpose: to determine the quantitative patterns and gradations of the degree of manifestation of changes in IRT indicators, depending on LDF data, corresponding to different conditions and peripheral blood flow disorders in the skin of laboratory rats. Material and Methods. Microcirculation of blood in bio-objects in background studies and immediately after the experimental modeling of hypobaric hypoxia was determined with the help of LDF, while simultaneously performing remote dynamic infrared thermography of the rats'tail skin. A comparison of LDF and IRT data was carried out using statistical methods of correlation-regression analysis. Results. The decrease in tissue blood flow was accompanied by a decrease in temperature, with an increase in perfusion of blood along the microcirculation pathways, the temperature of the skin surface increased. The quantitative criteria of experimental diagnostics in the form of boundary values and ranges of temperature indicators for an estimation of a different degree of severity of violations of the microcirculation of blood (peripheral hemodynamics) are determined. With a decrease or increase in tissue blood flow to 10, 10-25, 25-40 and more than 40% of the baseline (background control), the negative or positive skin surface temperature increase was up to 0.6, 0.6-1.2, 1.2-1.8 and more than 1.8°C respectively. Conclusion. Results of thermal imaging can be used as a criterion for predictive assessment of the state and changes in cutaneous peripheral blood flow in laboratory rats.

Detection of fibrous astrocytes on histological sections of hippocampi using two different methods of silver impregnation

Summary:

Цель: выделение клеток волокнистой астроцитарной глии на гистологических срезах гиппокампов при использовании двух различных растворов солей серебра. Материал и методы. Гистологические срезы гиппокампов кроликов, разделенные на две группы, импрегнировали растворами азотнокислого и бромистого серебра. Результаты. На гистологических срезах гиппокампов 1-й группы импрегнировались коротколучистые волокнистые астроциты (ВА). Около круглых ядер наблюдали различную плотность расположения глиофибрилл. Во 2-й группе импрегнировались длиннолучистые ВА с радиально расходящимися тонкими глиофибриллярными отростками. Заключение. Правильное представление об истинной структуре многоотростчатых клеток (содержащих глиофибриллы) можно получить при сравнительном изучении и анализе гистологических препаратов, окрашенных с помощью различных методов импрегнации солями серебра.

Microgliocyte argentation on histological sections of hippocampus of rabbits

Summary:

Purpose: to obtain morphological data to identify microglia on histological sections of hippocampus of rabbits using silver salt impregnation. Material and methods. Histological sections of hippocampus of rabbits impregnated with salt deposited silver. Using the Rio Hortega method for detecting fibrous glia, choose the desired chemical composition and acid-base parameters of the silver solution and reducing agent for the detection of microglia. Results. In histological sections of the hippocampus revealed microglia contrast with branched spikes and cells without shoots with thin flagellums. Conclusion. In visiting the experiments carried out in natural conditions, it is crucial to conduct biomedical research in the shortest possible time. We need to get not only the functional results of research, but also by their morphological, substantiating the data. The present method of impregnation microglia can be used as a rapid method for the morphological study of cerebral blood flow and neural tissue hypoxia.

Use of a remote infrared thermography in experimental medicine at extreme influences

Summary:

The study presents an analysis of published data on the use of remote infrared thermography in medicine and veterinary science and evaluated the main opportunities, methods and ways of thermovision registration in experimental medical studies on biological objects, including at extreme influences. The following resources of the bibliographic electronic databases had been used: eLibrary, PubMed, Clinical Key, Science. Now methods of infrared thermography in insufficient degree are used in experimental medicine, including when studying adverse biological effect of various harmful and dangerous extreme factors, which main manifestation, especially in the early period after exposure, are systemic microvascular disorders, determining the state of capacity of human biomodels. These dynamic remote ther-mographic studies of experimental biological objects can be used to evaluate disorders of the peripheral circulation, working capacity and an emotional condition of the biomodels exposed to extreme factors, including early after experimental studies in laboratory and field work.

Identify of argyrophilic cells of astrocytic glia to determine the hypoxic changes in nerve tissue

Summary:

Aim: identification of astrocytic glia cells impregnated to determine the hypoxic changes in nerve tissue. Material and methods. Condition of hypoxia was simulated in laboratory rats with "jar hypoxia". For the impregnation of histo-logical sections were used as a basis for the method of Rio-Gortega. Results. Silver impregnation method revealed several morphological forms of astrocytic cells in hypoxic changes in the nervous tissue of the brain. Conclusion. There were marked morphological brain astrocyte cell forms reflecting their functional state. Traced compensatory changes astrocytic cell depletion of these processes and the appearance of pathological morphological characters, up to the fragmentation and destruction of cells.

Morphometric parameters of air in lungs of laboratory rats on physical exercise.

Summary:

Aim: to determine the morphological and functional relationships between the morphometric parameters of lung airness tissue specimens of laboratory rats and indicators of physical endurance while swimming with weights and running on a treadmill. Material and methods. The physical endurance of laboratory rats evaluated in tests of swimming with weights and running on a treadmill using computer video markerless tracking. After strenuous exercise prepared histologic lung stained with hematoxylin and eosin. In morphometric study there were determined areal parameters of lung airness and pulmonary distelectasis. Results. When comparing the results of studies of physical endurance to the morphometric data of lung airness and pulmonary distelectasis identified the parameters of the same type of regression. Between changes in the morphological and physiological characteristics was a strong and average power correlation. The highest coefficient of correlation was established between the indicators of physical endurance and the parameter characterizing the degree of pulmonary distelectasis. Conclusion. Indicators of physical performance were the best in the big lung airness. At the same time, to a lesser extent than in the lung were presented wears off portions of the alveoli, the longer the biological objects to perform physical work. Fitted regression dependence characterizing the effect on physical endurance performance airiness of the lungs and spread them distelectasis.

Thermovision registration of defecation and urination in the evaluation of emotional state of experimental biological objects in the open arenas of behavioral tests

Summary:

Purpose: to provide a method for detection of boluses defecation and urination spots in the study of the behavior of experimental biological objects in open arenas ethological tests with thermal registration. Material and methods. For identifying and quantifying defecation and urination biological objects using the compact thermal imaging camera FLIR SC 660 (FLIR Systems) with a wide-angle lens. Object of study is the laboratory rat. Morphometric parameters determined using image analysis software. Results. When used as a video recording device thermal imaging camera to get an objective, quantitative data on the level of defecation and urination in the study of behavior and emotional state of the experimental evaluation of warm-blooded biological objects in open arenas ethological tests. Recorded boluses defecation and urination spots in the study of the emotional component of the behavior of laboratory rats in the test apparatus «Open field», «Elevated plus maze», «Radial 8-Arm Maze» and «T-maze». Conclusion. Thermal video registration allows visualizing products vegetative urinary and fecal excretion in the dynamics of experimental studies in the evaluation of the psycho-emotional state of biological objects in various open arenas behavioral tests, carried out their detailed morphometric analysis and identify indicators of spatial distribution.

Register indicators of physical endurance of biological objects when running a treadmill and swimming with weights using computer video markerless tracking

Summary:

Purpose: to study the use of video tracking to assess physical endurance and indicators of biological objects fatigue when running on a treadmill and swimming with the load. Material and methods. Physical endurance evaluated by test facilities for running on a treadmill and swimming with the load. As the object of the studies used laboratory rats. Results. For indicators of physical endurance biological objects isolated areas running track of treadmill and electrical stimulation site, when swimming on the total area of the container isolated subarea near the water surface. With video tracking performed computer timing of finding biological object in different zones of the treadmill and containers for swimming. On the basis of data on the time location rats in a given zone apparatus for running and swimming, obtained in the dynamics of the test of physical endurance, build a "fatigue curves", quantified changes in the indices of hard work, depending on the duration of its execution. Conclusion. Video tracking allows to define the execution of physical work to overflowing with loads of aerobic and mixed aerobic-anaerobic power, establish quantitative indicators of changes in the dynamics of biological objects operability testing with the construction of "fatigue curve" and objectively determine the times of occurrence in experimental animals exhaustion when fails to perform physical work.

Complex microscopic express-analysis for biomedical full-scale investigation

Summary:

To perform biomedical full-scale investigation there had been developed a set of microscopic analysis of histologi-cal preparations. This complex includes cryotome for histological sections, a set of chemical reagents and laboratory glassware for staining, binocular loupes for registration microstructural changes, a microscope with a camera and lighting system on swivel stand, modernized otoscope, a laptop with graphics programs. The complex provides quantitative indicators needed to diagnose the severity and probability estimates of the effect estimation of impacts of extreme factors of different nature.
